      研究目的:观察 双固一通 灸法对糖尿病及糖尿病周围神经病变大鼠血脂代谢、炎性因子分泌及肝脏组织中胰岛素受体(INSR)基因表达的影响。研究方法:大鼠分为正常组(A)、糖尿病模型组(B)、糖尿病模型艾灸治疗组(C)、糖尿病周围神经病变模型组(D)、糖尿病周围神经病变模型艾灸治疗组(E),于4周治疗结束后和8周治疗结束后空腹眼底静脉采血测各组大鼠血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),于治疗4周、8周后,用ELISA法测定血清肿瘤坏死因子TNF-α和白介素IL-6的含量,以及运用实时定量聚合酶链式反应方法检测肝脏胰岛素受体INSR的含量。研究结果:DM治疗组(C组)与DM模型组(B组)相比,TC、TG、LDL-C水平降低(P<0.05),HDL-C水平升高(P<0.05);DPN治疗组(E组)与DPN模型组(D组)相比,TC、TG、LDL-C水平降低(P<0.05),HDL-C水平升高(P<0.05)。DM模型组(B组)和DPN模型组(D组)血清肿瘤坏死因子TNF-α和白介素IL-6的含量显著高于正常(P<0.05);与B组比较,C组大鼠TNF-α和IL-6的含量均降低(P<0.05);与D组比较,E组大鼠TNF-α和IL-6的含量均降低(P<0.05)。DM模型组(B组)和DPN模型组(D组)肝脏胰岛素受体INSR表达明显低于正常(P<0.05);C组与B组相比,INSR表达明显增加(P<0.05);E组与D组相比,INSR表达明显增加(P<0.05)。研究结论:“双固一通”艾灸法治疗后,改善了糖尿病及周围神经病变大鼠血脂紊乱以及炎性因子的分泌的情况,也使得糖尿病和周围神经病变大鼠的胰岛素受体数量增加、功能增强,改变了胰岛素受体数量和功能缺陷所导致的胰岛素作用减弱的状态,进而改善胰岛素抵抗情况,这可能也是“双固一通”艾灸法防治糖尿病及糖尿病周围神经病变的作用机制之一。

      Objective: to observe the effect of " Shuanggu Yitong 、inflammatory factor secretion and insulin receptor (INSR) gene expression in diabetic and diabetic peripheral neuropathy rats.Research methods:Rats were divided into normal group (A), diabetes model (B), diabetes model moxibustion treatment group (C), diabetes peripheral neuropathy model group (D), diabetic peripheral neuropathy model moxibustion treatment group (E), in 4 weeks after treatment and 8 weeks after the treatment on an empty stomach fundus vein blood test groups of rats serum total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL - C), low density lipoprotein cholesterol (LDL - C), in 4 weeks, 8 weeks after treatment, with ELISA method of determination of serum TNF tumor necrosis factor alpha and interleukin 6 levels of IL -,Real-time quantitative polymerase chain reaction was used to detect the content of insulin receptor INSR in liver.Results: compared with the DM model group (group B), the levels of TC, TG and ldl-c in the DM treatment group (group C) were decreased (P<0.05), while the levels of hdl-c were increased (P<0.05).Compared with DPN model group (D group), the levels of TC, TG and ldl-c in DPN treatment group (E group) were decreased (P<0.05), while the levels of hdl-c were increased (P<0.05).The levels of TNF- and il-6 in serum of DM model group (group B) and DPN model group (group D) were significantly higher than normal (P<0.05).TNF- and il-6 levels in group C were lower than those in group B (P<0.05).The levels of TNF- and il-6 in group E were lower than those in group D (P<0.05).The expression of insulin receptor INSR in liver of DM model group (group B) and DPN model group (group D) was significantly lower than normal (P<0.05).INSR expression was significantly increased in group C compared with group B (P<0.05).INSR expression was significantly increased in group E compared with group D (P<0.05).Research conclusion: "double solid on the" method of moxibustion therapy, improved diabetes and peripheral neuropathy in rats blood lipid disorders and inflammation of the situation, also makes the diabetic peripheral neuropathy in rats and increase the number of insulin receptors, enhancements, and changes the number of insulin receptor and functional defects caused by insulin function weakened state, and thus improve the insulin resistance, it may also be a " Shuanggu Yitong
