      目的:通过观察针刺捻转补泻手法对自发性高血压大鼠(SHR)海马形态的改变、糖皮质激素受体(GR)表达及下丘脑-垂体-肾上腺皮质(HPA)轴各组分表达的影响,探讨其调控血压的中枢-外周机制。方法:48只雄性SHR大鼠随机等分为模型组、捻转补法组和捻转泻法组,16只WKY大鼠作为空白对照组。各针刺组分别采用不同的针刺手法干预双侧太冲穴,模型组和空白组不做针刺处理,各组于针刺前及针刺后定期测量血压。实验第16天采用HE染色法观察海马组织病理性改变,采用Elisa法检测海马GR及HPA轴各组分的表达。结果:与模型组比较,捻转泻法组和捻转补法组均能明显降低SHR大鼠血压(P<0.01,P<0.05),各针刺组海马GR含量明显降低(P<0.05),HPA轴各组分表达量明显降低(P<0.05);且捻转泻法组较捻转补法组降压效果和良性调控HPA轴作用更明显(P<0.05)。结论:针刺捻转补泻手法可以通过调节海马中GR含量表达,抑制HPA轴活动,起到降压作用,这可能是针刺捻转补泻手法降压的机制之一。
英文摘要:
      Objective:To observe the effects of twirling reinforcing-reduc ing manipulation(TRRM) on the changes in hippocampal morphology and the expression of glucocorticoid receptor(GR) and hypothalamic-pituitary- adrenal cortex (HPA) axis in spontaneously hypert ensive rats (SHR),and to explore the central-peripheral mechanism of blood pressure.Me thods:Forty-eight male SHR rats were randomly and equally divided into model group(M), twirling reinforcing group(B) and twirling reducing group(X) ,sixteen WKY rats as blank control group(K).The according acupuncture interventions at Taichong (LR3) acupoint were administered in the B and X groups. The rats in the K and M groups received no treatment,Each group regularly measured blood pressure before acupuncture and after acupuncture.On the 16th day of the experiment, the pathological changes of hippocampus were observed by HE staining, and the expression of GR in hippocampus and HPA axis components were detected by ELISA method. Results:Compared with the M group, the blood pressure showed a significant decrease in the X and B groups(P<0.01,P<0.05), the expression of the GR content of hippocampus and HPA axis components in each acupuncture group were significantly decreased (p<0.05).Furthermore, the X group showed the better effects of lowering the BP and benignantly regulating the HPA axis than that of other groups (P<0.05).Conclusion:The TTRM can regulate the expression of GR in the hippocampus to inhibit the activity of HPA axis, and play a role in blood pressure reduction,which may be one of the mechanisms of TTRM on antihypertensive effects.
