目的:探讨针刀对于膝骨关节炎(KOA)兔股直肌萎缩的修复作用。方法:雄兔随机分为针刀组(A)、电针组(EA)、正常组(N)和模型组(M),各9只。采用左后肢膝关节制动6周制备退行性膝骨关节炎模型。N组与M组不予干预,A组和EA组各干预4周。采用Western Blot法观测兔股直肌中Akt、mTOR、p70s6k的蛋白含量。结果:①M组较N组股直肌中mTOR和p70s6k、Akt蛋白的表达水平明显下降(P<0.01或P<0.05);②股直肌Akt蛋白表达:A组与EA组相比M组显著升高(P<0.01);③股直肌mTOR蛋白表达:相比M组,A组与EA组升高(P<0.01或P<0.05);④股直肌p70s6k蛋白表达:A组较M组显著升高(P<0.05),而EA组与M组无显著差异(P>0.05)。结论:针刀通过修复萎缩的股直肌,改善膝周软组织的生物力学,促进膝关节的恢复。 |

Objective: To investigate the repairing effect of Acupotomy on atrophy of rabbit rectus muscle in knee osteoarthritis (KOA). Methods: Male rabbits were randomly divided into acupuncture group (A), electroacupuncture group (EA), normal group (N) and model group (M), with 9 rabbits in each group. The knee osteoarthritis model was prepared by braking the left hind limb knee joint for 6 weeks. group N and group M were not intervened but group A and group EA were interfered for 4 weeks. The protein expression of Akt, mTOR and p70s6k in the rectus femoris muscle was detected by Western Blot. Results:① The expression levels of mTOR, p70s6k and Akt in the rectus femoris of group M were significantly lower than those in group N (P<0.01 or P<0.05). ②The expression of Akt protein in rectus muscles : group A and group EA was significantly higher than those in group M (P<0.01);③mTOR protein expression in rectus muscle: compared with group M, group A and EA group increased (P<0.01 or P<0.05); ④p70s6k protein expression in rectus muscle: There was a significant increase in the group A compared with group M (P<0.05), but there was no significant difference between the EA group and the M group (P>0.05). Conclusion: Acupotomy can promote the recovery of knee joint by repairing rectus femoris and improving the biomechanics of knee peri-soft tissue. |
