      目的:研究针灸对骨骼肌萎缩模型大鼠的干预效果及作用机制。方法:30只大鼠随机分为模型组、假手术组和针灸组,各10只,建立骨骼肌萎缩模型。针灸组给予针灸治疗,假手术组和模型组大鼠每天进行固定。检查腓肠肌湿重、干重,骨骼肌肌纤维直径、面积,采用ELISA检测泛素蛋白连接酶肌肉环指蛋白1(MURF1)、PGC-1α、Ⅲ型纤连蛋白组件包含蛋白5(FNDC5)蛋白,采用Western Blot检测Akt、mTOR、p-Akt、p-mTOR。结果:针灸组骨前肌湿重、干重、干重/体重、骨骼肌肌纤维直径、面积高于模型组,低于假手术组(P<0.05)。模型组骨前肌湿重、干重、干重/体重、骨骼肌肌纤维直径、面积低于假手术组(P<0.05)。针灸组骨骼肌相关蛋白MURF1表达低于模型组,PGC-1α、FNDC5表达高于模型组;针灸组骨骼肌相关蛋白MURF1表达高于假手术组,PGC-1α、FNDC5表达低于假手术组(P<0.05)。模型组骨骼肌相关蛋白MURF1表达高于假手术组,PGC-1α、FNDC5表达低于假手术组(P<0.05)。针灸组Akt、mTOR、p-Akt、p-mTOR表达高于假手术组和模型组(P<0.05)。模型组Akt、mTOR、p-Akt、p-mTOR表达低于假手术组(P<0.05)。结论:针灸对骨骼肌萎缩模型大鼠干预效果显著,能改善大鼠骨骼肌萎缩状况,通过激活Akt/mTOR通路,调控骨骼肌相关蛋白MURF1、PGC-1α、FNDC5表达,从而抑制骨骼肌萎缩。

      Objective:To study the intervention effect and mechanism of acupuncture and moxibustion on skeletal muscle atrophy model rats.Methods:Thirty rats were randomly divided into model group, sham-operated group and acupuncture group, with 10 rats in each group. Skeletal muscle atrophy model was established. Acupuncture and moxibustion were given in the acupuncture group, while sham operation group and model group were fixed daily. Wet and dry weight of gastrocnemius muscle, diameter and area of skeletal muscle fibers were examined. Ubiquitin protein ligase muscle ring finger protein 1 (MURF1), PGC-1α and type III fibronectin components containing protein 5 (FNDC5) were detected by ELISA. Akt, mTOR, p-Akt and p-mTOR were detected by Western Blot.Results:The wet weight, dry weight, dry weight/weight, diameter and area of skeletal muscle fibers in the acupuncture group were higher than those in the model group and lower than those in the sham operation group (P<0.05). The wet weight, dry weight, dry weight/weight, diameter and area of skeletal muscle fibers in model group were lower than those in sham operation group (P<0.05). The expression of MURF1 was lower in the acupuncture group than in the model group, and the expressions of PGC-1α and FNDC5 were higher in the acupuncture group than in the model group. The expression of MURF1 was higher in the acupuncture group than in the sham operation group, while the expressions of PGC-1α and FNDC5 were lower in the acupuncture group than in the sham operation group (P<0.05). The expression of MURF1 in model group was higher than that in sham-operated group, and the expression of PGC-1α and FNDC5 was lower than that in sham-operated group (P<0.05). The expressions of Akt, mTOR, p-Akt and p-mTOR in acupuncture group were higher than those in sham operation group and model group (P<0.05). The expression of Akt, mTOR, p-Akt and p-mTOR in model group was lower than that in sham operation group (P<0.05).Conclusion:Acupuncture and moxibustion can improve the condition of skeletal muscle atrophy in rats. By activating the Akt/mTOR pathway, it can regulate the expression of MURF1, PGC-1α and FNDC5 in skeletal muscle, thereby inhibiting skeletal muscle atrophy.
