      目的:观察电针对慢性应激抑郁模型大鼠中缝核中P11、tPA、BDNF的影响,进一步研究电针治疗抑郁症可能的通路。方法:将24只健康雄性SD大鼠随机分为正常组、模型组、药物组、电针组,每组6只。除正常组外,其余各组均孤养,并接受慢性不可预知应激刺激造模。各组大鼠分别于实验前1天和实验的第28天进行体质量检测和糖水偏好实验,观察大鼠的行为学变化。实验第28天断头取脑,采用逆转录酶聚合酶链反应检测中缝核中P11、tPA、BDNF mRNA的含量;免疫荧光染色技术中缝核中三者的表达情况。结果:模型组大鼠的行为学指标明显低于正常组(P<0.05),药物组和电针组大鼠行为学指标均明显高于模型组(P<0.05);模型组中缝核P11、tPA、BDNF的mRNA含量低于正常组(P<0.01),药物组和电针组则显著高于模型组(P<0.01);模型组免疫荧光染色结果显示模型组三者在中缝核的表达明显低于正常组,药物组和电针组明显高于模型组。结论:电针有明显的抗抑郁效果,可以改善慢性应激抑郁大鼠的行为;电针可以增加慢性应激抑郁大鼠模型中缝核P11、tPA、BDNF mRNA的含量,提高其表达量,电针的这种抗抑郁作用可能是通过影响P11-tPA-BDNF通路发挥作用的。

      Objective: To observe the effects of electroacupuncture on P11, tPA and BDNF in the nucleus of rats with chronic stress depression, and to further study the possible pathways of electroacupuncture for depression. Methods: Twenty-four healthy male Sprague-Dawley rats were randomly divided into normal group, model group, drug group and electroacupuncture group, with 6 rats in each group. Except the normal group, the other groups were orphaned and subjected to chronic unpredictable stress stimulation modeling. The body mass test and the syrup preference test were performed on the rats 1 day before the experiment and the 28th day of the experiment, and the behavioral changes of the rats were observed. On the 28th day of the experiment, the brain was decapitated, and the contents of P11, tPA and BDNF mRNA in the nucleus raphe were detected by reverse transcriptase polymerase chain reaction; the expression of the three in the nucleus was detected by immunofluorescence staining. Results: The behavioral indexes of the model group were significantly lower than those of the normal group (P<0.05). The behavioral indexes of the rats in the drug group and the electroacupuncture group were significantly higher than those in the model group (P<0.05). The mRNA levels of tPA and BDNF were lower than those in the normal group (P<0.01), and the drug group and electroacupuncture group were significantly higher than the model group (P<0.01). The immunofluorescence staining results of the model group showed the expression of the model group in the nucleus raphe Significantly lower than the normal group, the drug group and the electroacupuncture group were significantly higher than the model group. Conclusion: Electroacupuncture has obvious antidepressant effect and can improve the behavior of rats with chronic stress depression. Electroacupuncture can increase the content of P11, tPA and BDNF mRNA in the rat model of chronic stress depression, and increase its expression. This antidepressant effect of the needle may be through the effects of the P11-tPA-BDNF pathway.
