      摘要: 目的:观察电针结合运动训练对心肌缺血再灌注损伤大鼠心肌细胞凋亡的影响。 方法:随机将60只SD大鼠分为对照组、模型组、电针组、运动组及电针结合运动训练组,共5组(n=12)。电针及运动训练干预3周后采用Langendorff灌流系统制作离体MIRI模型。TTC染色法检测各组大鼠心肌梗死面积,TUNEL染色法检测心肌细胞凋亡;Western Blot检测心肌组织中Bax、Bcl-2的蛋白表达。 结果:TTC染色结果显示:与对照组比较,模型组心肌梗死范围增大(P<0.05);与模型组比较,其余各干预组心肌梗死范围缩小(P<0.05);与电针组、运动组相比,联合组心肌梗死范围缩小(P<0.05);电针组和运动组比较,差异无统计学意义(P>0.05)。TUNEL阳性细胞比值结果:对照组见极少数TUNEL阳性细胞。与对照组比较,模型组TUNEL阳性细胞比值明显增加(P<0.05);与模型组比较,电针组、运动组TUNEL阳性细胞比值明显减少(P<0.05);与电针组、运动组比较,联合组TUNEL阳性细胞比值明显增加(P<0.05);电针组和运动比较,差异无统计学意义(P>0.05)。WB结果:各组心肌组织Bax、Bcl-2蛋白表达:Bax蛋白的表达:与对照组比较,模型组Bax蛋白表达升高(P<0.05);与模型组比较,电针组、运动组Bax蛋白表达降低(P<0.05);与电针组、运动组比较,联合组Bax蛋白表达降低(P<0.05);电针组和运动组比较,差异无统计学意义(P>0.05)。Bcl-2蛋白表达:与对照组比较,模型组Bcl-2蛋白表达降低(P<0.05);与模型组比较,电针组、运动组Bcl-2蛋白表达升高(P<0.05);与电针组、运动组比较,联合组Bcl-2蛋白表达升高(P<0.05);电针组运动组比较,差异无统计学意义(P>0.05)

      Summary: Objective: To observe the effect of electroacupuncture combined with exercise training on cardiomyocyte apoptosis in rats with myocardial ischemia-reperfusion injury. Methods: 60 SD rats were randomly divided into 5 groups: control group, model group, electroacupuncture group, exercise group and electroacupuncture combined with exercise training group (n = 12). After 3 weeks of electroacupuncture and exercise training intervention, Langendorff perfusion system was used to make in vitro MIRI model. TTC staining was used to detect myocardial infarction area and TUNEL staining was used to detect cardiomyocyte apoptosis; The protein expressions of Bax and Bcl-2 in myocardial tissue were detected by Western blot. Results: TTC staining showed that compared with the control group, the scope of myocardial infarction in the model group increased (P < 0.05); Compared with the model group, the scope of myocardial infarction in other intervention groups was reduced (P < 0.05); Compared with electroacupuncture group and exercise group, the scope of myocardial infarction in the combined group was reduced (P < 0.05); There was no significant difference between electroacupuncture group and exercise group (P > 0.05). Results of TUNEL positive cell ratio: very few TUNEL positive cells were found in the control group. Compared with the control group, the ratio of TUNEL positive cells in the model group increased significantly (P < 0.05); Compared with the model group, the ratio of TUNEL positive cells in electroacupuncture group and exercise group decreased significantly (P < 0.05); Compared with electroacupuncture group and exercise group, the ratio of TUNEL positive cells in the combined group increased significantly (P < 0.05); There was no significant difference between electroacupuncture group and exercise group (P > 0.05). WB results: the expression of Bax and bcl-2 protein in myocardial tissue of each group: the expression of Bax protein in model group was higher than that in control group (P < 0.05); Compared with model group, the expression of Bax protein in electroacupuncture group and exercise group decreased (P < 0.05); The expression of Bax protein in the combined acupuncture group was lower than that in the exercise group (P < 0.05); There was no significant difference between electroacupuncture group and exercise group (P > 0.05). Bcl-2 protein expression: compared with the control group, the bcl-2 protein expression in the model group decreased (P < 0.05); Compared with the model group, the expression of Bcl-2 protein in electroacupuncture group and exercise group increased (P < 0.05); Compared with electroacupuncture group and exercise group, the expression of Bcl-2 protein in combined group increased (P < 0.05); There was no significant difference between electroacupuncture group and exercise group (P > 0.05).
